Isolation and Separation of Epidermal and Mesophyll Protoplasts from Rye Primary Leaves — Tissue-Specific Characteristics of Secondary Phenolic Product Accumulation

چکیده
We have developed a technique for the large-scale isolation of epidermal and m esophyll proto plasts, as well as the vascular strands, o f rye primary leaf blades. Separation o f the two types of protoplasts has been successful only from leaves harvested at the end of a 13-h light period, when chloroplasts were enriched in starch. The occurrence o f different flavonoid compounds, and amounts, in epidermal and m esophyll protoplasts can be used as criteria for protoplast purity and viability since C-glucosylflavone Oglycosides are characteristic of epidermal protoplasts whereas flavone O-glucuronides and anthocyanins are typical o f mesophyll protoplasts. Several non-flavonoid phenolic com pounds are found only in the epidermal protoplast. These patterns of secondary product accumulation reflect the high tissue specificity of the rye leaf.

PLANT REGENERATION FROM MESOPHYLL PROTOPLASTS OF POTATO (SOLANUM TUBEROSUM L.) CULTIVAR DELAWARE USING SILVER THIOSULFATE (STS)
The use of in vitro potato shoot cultures for protoplast isolation is desirable for consistent protoplast qualities. Ethylene build-up in the culture vessels causes problems for leaf growth (e.g. decreasing leaf surface, dry weight and chlorophyll) in shoot culture. Fifty to one hundred µM Silver thiosulfate (STS) produced a larger leaf area as a source of plant material for protoplast isolatio...
متن کاملDifferential ion accumulation and ion fluxes in the mesophyll and epidermis of barley.
In barley (Hordeum vulgare L.) leaves, differential ion accumulation commonly results in inorganic phosphate (Pi) being confined to the mesophyll and Ca(2+) to the epidermis, with preferential epidermal accumulation of Cl(-), Na(+), and some other ions. The pattern was confirmed in this study for major inorganic anions and cations by analysis of barley leaf protoplasts. The work focused on the ...
متن کاملCadmium uptake and sequestration kinetics in individual leaf cell protoplasts of the Cd/Zn hyperaccumulator Thlaspi caerulescens.
Hyperaccumulators store accumulated metals in the vacuoles of large leaf epidermal cells (storage cells). For investigating cadmium uptake, we incubated protoplasts obtained from leaves of Thlaspi caerulescens (Ganges ecotype) with a Cd-specific fluorescent dye. A fluorescence kinetic microscope was used for selectively measuring Cd-uptake and photosynthesis in different cell types, so that phy...
متن کاملCyanidin 3-Gentiobioside from Primary Leaves of Rye (Secale cereale L.)
Primary leaves of rye (Secale cereale L. cv. Kustro) show a heterogeneous, tissue-specific pattern of flavonoids being composed of the two C-glycosylflavone glycosides isovitexin 2"-0-arabinoside and isovitexin 2"-0-galactoside (located in the epidermal layers) [1, 2], the two flavone glycosides luteolin 7O-diglucuronide and luteolin 7-O-diglucuronide 4'O-glucuronide [3] and the two anthocyanin...
متن کاملTissue Distribution and Subcellular Localization of Prephenate Aminotransferase in Leaves of Sorghum bicolor.
The tissue and subcellular distribution of prephenate aminotransferase, an enzyme of the shikimate pathway, was investigated in protoplasts from leaves of Sorghum bicolor. Activity was detected in purified epidermal and mesophyll protoplasts, and in bundle sheath strands. After fractionation of mesophyll and epidermal protoplasts by differential centrifugation, 92% of the total prephenate amino...
